Testosterone is a secreted glycoprotein, which was originally identified from the enriched culture medium of sertoli cells in mice. It is a cytoskeleton-related protein with small molecular weight, which can be closely bound to the cell surface through receptor-related proteins.
Decision principle
Radioimmunoassay: Testosterone is mainly produced in interstitial cells between seminiferous tubules of testis, and can also be synthesized in adrenal cortex. When h testosterone and testosterone antibody are added to the sample, competition inhibition will occur.
In the reaction system, when the amount of H testosterone and its antibody remains constant, the amount of labeled antibody complex is limited by the amount of unlabeled antigen. Dextran-coated activated carbon is used for separation, so that the bound labeled antigen is separated from the free labeled antigen. Determine the binding part of the precipitate, and calculate the content of the unknown sample by comparing the liquid scintillation counting (dpm) with the standard.
