Antithrombotic effect of glycosaminoglycan from sea cucumber;

Sea cucumber glycosaminoglycan (hGAG) is a kind of glycosaminoglycan containing fucose (L-fu cos) extracted from the sea cucumber body wall widely grown in the South China Sea. In the early 1980s, when Chinese scholars studied its anti-tumor effect, they found that hGAG could cause bleeding in experimental animals, and people gradually realized its anticoagulant effect. The researchers used hGAG to treat normal people and patients with cardiovascular and cerebrovascular thrombotic diseases, and found that HGAG has the functions of anticoagulation, lowering blood viscosity and lowering blood lipid. In order to better utilize and develop sea cucumber, a marine biological resource, the researchers studied the antithrombotic mechanism of hGAG. Methods: The effects of hGAG on the procoagulant activity, tissue factor (TF), thrombomodulin (TM) and plasminogen activator inhibitor-1(PAI- 1) of endothelial cells were observed. Results: After bacterial lipopolysaccharide (LPS) was incubated with endothelial cells, different concentrations of hGAG could reduce procoagulant activity, TF antigen and mRNA transcription, and enhance TM antigen expression and mRNA transcription. HGAG can decrease the antigen, activity and mRNA transcription of PAI- 1 in the supernatant of endothelial cells. Conclusion: hGAG plays an antithrombotic role by down-regulating TF expression in endothelial cells, promoting TM expression, reducing the synthesis and secretion of PAI- 1 and inhibiting the transcription of PAI- 1mRNA.
