Among them, in biochemical experiments, many experiments related to protein and nucleic acid need to use Tris as biological buffer. There is even a tendency to exceed phosphate buffer, because it has good buffering capacity, low toxicity, extremely low interference to biological reactions and high purity. There is a tendency to exceed phosphate. For example, TRIS buffer has been tested in SDS- polyacrylamide gel electrophoresis, and phosphate is rarely used.
Application of tris (trimethylaminomethane)
① The buffer system composed of glycine and electrophoresis buffer is used to stabilize the P value;
(2) using TRIS-HCL buffer system to stabilize the PH value in the gel;
③ Widely used as a solvent for nucleic acid and protein;
The low ionic strength of TRIS buffer can also be used to form intermediate fibers of nematodes;
⑤ Adding EDTA into TRIS hydrochloric acid buffer to make "TE buffer", which can be used to stabilize and preserve DNA;
⑥ Use acetic acid instead of acidic solution to get "TAE buffer", and use boric acid instead to get TBE buffer. These two solutions are commonly used in nucleic acid electrophoresis.
